Identification and Clarification of VrCYCA1: A Key Genic Male Sterility-Related Gene in Mungbean by Multi-Omics Analysis

Heterosis has been utilized in crops for a long time, and although crop fertility is the basis utilization of heterosis, there limited information concerning genic male sterility (GMS) mungbean. Therefore, based on sterile mutant, M1, obtained by EMS mutagenesis Weilyu cultivar, this project used multi-omics analysis to detect site further identify its candidate gene, then mechanism gene was discussed. As result, one locus region (Chr5: 6,835,001–6,935,000 bp) associated with GMS identified, nine genes were found within 100 Kb region. The VrCYCA1, around above loci had TATA box deletion approximately 4.7 upstream evidenced comparative genomics, transcriptome analysis, RT-qPCR analysis. expression level VrCYCA1 significantly downregulated (log2FC = −2.06, p-value 0.025) ms lines compared control group. Moreover, 6653 showed differential between mutant as well 165 metabolites significant differences their concentration levels. Among those differentially expresses genes, 226 annotated functional categories involved flowering endosperm development, six protein–protein interactions VrCYCA1. Seven seven participated relationship reproductive growth vegetative growth, which might have caused mungbean plants. This study data mine GMS-related constructed genetic network explore molecular results lay solid foundation biology research sterility.
